Saizen available forms :
Injection : 5 mg (about 15 international units / vial)
Saizen indications :
  • Saizen (Somatropin) is used to treat : growth failure in children due to chronic renal insufficiency, growth failure in children due to deficiency of growth hormone, short stature associated with Turner's syndrome.
Saizen action :
  • Saizen produces growth (skeletal and cellular).Metabolic actions include : increased protein synthesis, increased carbohydrate metabolism, lipid mobilization, retention of sodium and potassium.Saizen has the same amino acid sequence as naturally occurring growth hormone.Growth hormone enhances gastrointestinal tract mucosal transport of water, electrolytes and nutrients.
  • Therapeutic effects of Saizen : increased skeletal growth in children with growth hormone deficiency, replacement of somatropin in deficient adults, decreased wasting in persons with AIDS, increased bone density in adult growth hormone, enhanced gastric absorption of water and nutrients in short bowel syndrome.

Saizen contraindications and precautions :
  • Saizen (Somatropin) is contraindicated in : closure of epiphyses, active neoplasia, hypersensitivity to growth hormone, acute critical illness or respiratory failure, diabetic retinopathy, Prader-Willi syndrome with obesity and respiratory impairment.
  • Use Saizen cautiously in : growth hormone deficiency due to intracranial lesion, coexisting adrenocorticotropic hormone (ACTH) deficiency, diabetes, geriatric persons, thyroid dysfunction, pregnancy, lactation.
Saizen adverse reactions and side effects :
  • Saizen side effects and adverse reactions can be : edema of the hands and feet, hyperglycemia, hypothyroidism, insulin resistance, pain at injection site, arthralgia.
Saizen dosage :

Subcut and intramuscular administration (children) :

  • 0.05 mg (0.15 unit / kg) 3 times weekly.
How to administer Saizen :
  • Reconstitute each 5-mg vial of Saizen (Somatropin) with 1 - 3 ml of bacteriostatic water for injection.Reconstituted Saizen vials are stable for 14 days if refrigerated.To use cool click needle free injector, wind the device to energize the spring, and draw Saizen into the Crystal Check nozzle.Using firm pressure at the injection site, hold the injector at a 90° angle and press the blue actuator button.
  • Rotate injection sites with each injection.
  • For subcut administration, injection volume for Saizen shouldn't exceed 1 ml.
Useful advices on Saizen :
  • Parents : learn the correct procedure for recontituting medication, site selection, technique for intramuscular or subcut injection, and disposal of needles and syringes.Review dosage schedule and report persistent pain or edema at injection site.
  • Administration of Saizen to persons without growth hormone deficiency or after epiphyseal closure may result in acromegaly (coarsening of facial features; enlarged hands, feet and internal organs; increased blood glucose; hypertension).
  • You MUST follow-up with endocrinologist to ensure appropriate growth rate, to evalute lab work and to determine bone age by X-Ray exam.
  • Saizen dose is synthetic and therefore is not capable of transmitting Creutzfeldt-Jakob disease, as was the original somatropin which was extracted from human cadavers.
  • Child with endocrine disorders (including growth hormone deficiency) may have an increased risk of slipped capital epiphyses.Notify prescriber immediately if you're notice that your child is limping.
  • Diabetics should monitor glucose level closely and report changes to prescriber.
  • Treatment may continue for years because regular assessments are required.
  • Saizen treatment is very expensive.
